The data generated by study of tonsil and appendectomy samples have also been revealing (Ironside et al., 2000; Hilton et al., 2002; Frosh et al., 2004). The first of the studies revealed three out of 12,674 samples positive for abnormal prion protein (PrP) on Western blot (Hilton et al., 2004a) where the specificity is thought to be high but the sensitivity uncertain (Hilton et al., 2004b).

The studies on frozen or paraffin-embedded tonsil tissues did not bring additional useful data for the estimation of prevalence of healthy carriers in the United Kingdom (Frosh et al., 2004; Clewley et al., 2009; de Marco et al., 2010). The genotype at codon 129 of the PRNP gene could be determined for 2 out of 3 patients of the first study (all valine-valine) and for all the patients of the second study (8 methionine-methionine, 4 methionine-valine, and 4 valine-valine).
