This chapter explains the maintenance of Xenopus laevis and oocyte injection. There are six species of Xenopus, all indigenous to Africa, but Xenopus laevis is the only one commonly used in laboratory research. To synchronize oocyte development, the frogs can be injected with human chorionic gonadotropin (hCG), which induces ovulation and egg laying, after which oocyte development starts over approximately synchronously. Xenopus can tolerate a wide range of temperature fluctuations, but the quality of oocytes diminishes markedly following temperature shifts. Therefore, frogs should be housed in a temperature-controlled environment. Xenopus are very susceptible to desiccation, and they can die from being out of the water for a few hours. Oocytes from Xenopus laevis can be classified into six stages of development on the basis of anatomy; the choice of which stage oocytes to inject depends on the purposes of the experiment.

Engineering voltage sensing phosphatase (VSP)

Voltage sensing phosphatase (VSP), consists of a voltage sensor domain (VSD) like that found in voltage-gated ion channels and a phosphoinositide (PIP) phosphatase region exhibiting remarkable structural similarity to a tumor suppressor enzyme, PTEN. Membrane depolarization activates the enzyme activity through tight coupling between the VSD and enzyme region. The VSD of VSP has a unique nature; it is a self-contained module that can be transferred to other proteins, conferring voltage sensitivity. Thanks to this nature, numerous versions of gene-encoded voltage indicators (GEVIs) have been developed through combination of a fluorescent protein with the VSD of VSP. In addition, VSP itself can also serve as a tool to alter PIP levels in cells. Cellular levels of PIPs, PI(4,5)P₂ in particular, can be acutely and transiently reduced using a simple voltage protocol after heterologous expression of VSP. Recent progress in our understanding of the molecular structure and mechanisms underlying VSP facilitates optimization of its molecular properties for its use as a molecular tool.
Effect of OBPs on the response of olfactory receptors

Aliquots of 1 μL are stored at − 80 °C. The protocol for frog dissection is taken from Goldin (1992) and Goldin and Sumikawa (1992) and illustrated in Fig. 1. Put an adult female African clawed frog into 1 L anesthetizing solution and cover with a layer of ice in a 5 L beaker.
The technique of two-electrode voltage-clamp (TEVC) recording from the heterologous expression system of olfactory receptors (ORs) in Xenopus laevis oocytes has been widely used to deorphanize insect ORs, that is to identify specific ligands for each of them. However, there is a controversial issue on whether ORs are activated by the odorant/OBP complex or the odorant alone. The mechanism of interaction among odorants, odorant-binding proteins (OBPs) and ORs remains largely unknown, due to the limitations in the use of scientific and innovative methods. In this chapter, the modified Xenopus oocytes expression system combined with TEVC technique is used to approach this issue. We describe the experimental strategies and provide detailed protocols for recording the signals generated by ORs in response to odorant/OBP complex at different concentrations. Results obtained by this approach have revealed that the presence of OBPs in the system affects the selectivity and sensitivity responses of ORs. Such studies help understanding the molecular mechanism of odorant detection in peripheral nervous system.
Induction of divalent cation permeability by heterologous expression of a voltage sensor domain

The voltage sensor domain (VSD) is a protein domain that confers sensitivity to membrane potential in voltage-gated ion channels as well as the voltage-sensing phosphatase. Although VSDs have long been considered to function as regulatory units acting on adjacent effectors, recent studies have revealed the existence of direct ion permeation paths in some mutated VSDs and in the voltage-gated proton channel. In this study, we show that calcium currents are evoked upon membrane hyperpolarization in cells expressing a VSD derived from an ascidian voltage-gated ion channel superfamily. Unlike the previously reported omega-pore in the Shaker K⁺ channel and rNav1.4, mutations are not required. From electrophysiological experiments in heterologous expression systems, we found that the conductance is directly mediated by the VSD itself and is carried by both monovalent and divalent cations. This is the first report of divalent cation permeation through a VSD-like structure.
Electrophysiological Analysis of Voltage-Gated Ion Channel Modulation by General Anesthetics

Voltage-gated ion channels (VGICs) of excitable tissues are emerging as targets likely involved in both the therapeutic and toxic effects of inhaled and intravenous general anesthetics. Whereas sevoflurane and propofol inhibit voltage-gated Na⁺ channels (Navs), sevoflurane potentiates certain voltage-gated K⁺ channels (Kvs). The combination of these effects would dampen neural excitability and, therefore, might contribute to the clinical endpoints of general anesthesia. As the body of work regarding the interaction of general anesthetics with VGICs continues to grow, a multidisciplinary approach involving functional, biochemical, structural, and computational techniques, many of which are detailed in other chapters, has increasingly become necessary to solve the molecular mechanism of general anesthetic action on VGICs. Here, we focus on electrophysiological and modeling approaches and methodologies to describe how our work has elucidated the biophysical basis of the inhibition Navs by propofol and the potentiation of Kvs by sevoflurane.

Xenopus laevis, also known as the African clawed frog, has played a significant role in understanding the causes of epilepsy and in identifying and characterizing effective treatments. This has largely been accomplished through the use of the oocytes of Xenopus, which possess many qualities that make them well-suited for the study of ion channels, neurotransmitter receptors, and membrane transporters involved in the generation of epileptic seizures. The Xenopus tadpole has also been used in important toxicology studies of antiepileptic drugs and as a model of chemically-induced seizures. In this chapter, the use of Xenopus as a model in the study of epilepsy and seizures will be detailed, as well as the advantages and limitations of its use.
The muscarinic antagonists scopolamine and atropine are competitive antagonists at 5-HT<inf>3</inf> receptors

Scopolamine is a high affinity muscarinic antagonist that is used for the prevention of post-operative nausea and vomiting. 5-HT₃ receptor antagonists are used for the same purpose and are structurally related to scopolamine. To examine whether 5-HT₃ receptors are affected by scopolamine we examined the effects of this drug on the electrophysiological and ligand binding properties of 5-HT₃A receptors expressed in Xenopus oocytes and HEK293 cells, respectively. 5-HT₃ receptor-responses were reversibly inhibited by scopolamine with an IC₅₀ of 2.09 μM. Competitive antagonism was shown by Schild plot (pA₂ = 5.02) and by competition with the 5-HT₃ receptor antagonists [³H]granisetron (K_i = 6.76 μM) and G-FL (K_i = 4.90 μM). The related molecule, atropine, similarly inhibited 5-HT evoked responses in oocytes with an IC₅₀ of 1.74 μM, and competed with G-FL with a K_i of 7.94 μM. The reverse experiment revealed that granisetron also competitively bound to muscarinic receptors (K_i = 6.5 μM). In behavioural studies scopolamine is used to block muscarinic receptors and induce a cognitive deficit, and centrally administered concentrations can exceed the IC₅₀ values found here. It is therefore possible that 5-HT₃ receptors are also inhibited. Studies that utilise higher concentrations of scopolamine should be mindful of these potential off-target effects.
